| Overview | Caspase-1(alsoknownasICE,Interleukin-1beta-ConvertingEnzyme)istheprototypicalmemberofthecaspasefamilyofcysteineproteases.Caspase-1existsincellsasaninactive45kDapro-enzyme.Thepro-enzymeismaturedbyproteolysistoyieldlarge(20kD)andsmall(10kD)subunits.Theactiveenzymeisaheterotetramerconsistingoftwolargeandtwosmallsubunits.Todatethemechanismofregulationofcaspase-1activationiscomplexandpoorlyunderstood.InTHP-1cells,alargeproportionofthecaspase-1ispresentintheinactivepro-enzymeform. Theactiverecombinanthumancaspase-1wasexpressedinE.coli.Theexpressedcaspase-1spontaneouslyundergoesautoprocessingtoyieldthesubunitscharacteristicofthenativeenzyme.Therecombinantcaspase-1preferentiallycleavesthesubstratesconsistingofconsensussequenceYVAD(e.g.,YVAD-AFCandYVAD-pNA).Therecombinantenzymecanalsobeusedasapositivecontrolincaspaseassaysorindeterminingthespecificityofcaspasesubstrates. Recommendedusage:0.5-1unitforfluorometriccaspaseassaysand2-4unitsforcolorimetriccaspaseassays.Optimalworkingdilutionsmustbedeterminedbyenduser. |
