| Overview | E-cadherinisacelladhesionproteinthatsuppressestumorcellinvasionandmetastasisintumormodels.OnmechanismregulatingtheexpressionoftheE-cadheringeneismethylationofthepromoterregion.TheCpGWIZ®E-cahedrinAmplificationKitisusedfordeterminingthemethylationofstatusoftheE-cadherinpromoterbymethylation-specificPCR(MSP).Thekitcontainsprimerstargetedtoregionsofthepromoterwherethesequencesaremostdivergentafterbisulfitetreatment.PCRparametershavebeenidentifiedsothatallprimersetsinthekitamplifyunderthesameconditions.ControlgenomicDNAsamples(methylatedandunmethylated)forE-cadherinarealsoincluded. Methylation-specificPCR(MSP)isanewtechnologyforsensitivedetectionofabnormalgenemethylationutilizingsmallamountsofDNA(Herman,1996).ThisprocessemploysaninitialbisulfitereactiontomodifytheDNA,followedbyPCRamplificationwithspecificprimersdesignedtodistinguishmethylatedfromunmethylatedDNA. |
