OverviewDESCRIPTION/COMPOSITION:CC117containsamixtureoflarge,extracellularchondroitinsulfateproteoglycansisolatedfromembryonicchickenbrain.Aminoacidsequenceanalysesandimmunologicalstudieshavedemonstratedthatthemajorcomponentsofthismixtureareneurocan,phosphacan,versican,andaggrecan.Followingchondroitinasetreatmenttoremovetheglycosaminoglycanportionoftheseproteoglycans,themajorcomponentsofthispreparationresolvedonSDSgelsare250kDacoreproteinsrepresentingneurocanand400kDacoreproteinsrepresentingaggrecan,phosphacan,andversican.

BackgroundInformationAggrecan,versican,andneurocanaremembersofthefamilyofhyaluronicacid-bindingproteoglycansandthereforeeachcontainshyaluronicacidbindingregions,Ig-likedomains,EGFlikedomains,lectin-likedomains,andcomplementregulatory-likedomains.Phosphacanisdistinct;itcontainsaregionofsimilaritytocarbonicanhydrase,afibronectintypeIIIdomain,andisasplicevariantofthetransmembraneproteinreceptortyrosinephosphataseequivalenttoitsextracellularportion.

AlthoughaggrecanwastrADItionallythoughtofasacartilageproteoglycanandneurocanandphosphacanwerethoughtofasneuralproteoglycans,recentstudiesindicatethatallfouroftheseproteoglycanshavebroadyetdistinctivedistributionpatternsinearlyembryos.Thepresenceoftheseproteoglycanshasnotbeencarefullyexaminedinadulttissuebeyondtheexpectedsites.Theseproteoglycansareanti-adhesionmoleculesandarecapableofinhibitingbothcell-ECMandcell-celladhesion.
